Although plasmacytoid dendritic cells (pDC) have been reported to be increased in several chronic inflammatory diseases, their role in allergic rhinitis (AR) remains unknown. Here, we demonstrate an increased number of circulating and tissue pDC, particularly in the sinonasal mucosa, in AR patients. Because these cells are known to drive the development of Th2 responses, the possibility of an involvement of pDC in AR was investigated by using well-characterized in vivo models of allergic rhinitis, nasal polyps, and rhinovirus infection. In vitro, the addition of interferon-alpha (IFN-alpha) to pDC, which inhibited the expression of interleukin-12, clearly inhibited the production of the Th2-associated cytokines, interleukin-4 and interleukin-5. To characterize the role of pDC in the allergic reaction in vivo, pDC were depleted in the nasal mucosa and in peripheral blood of immunized mice. This resulted in a significant decrease in allergic responses, such as eosinophilic inflammation, cytokine production, and IgE production. Finally, the application of an IFN-alpha-containing nasal spray resulted in an inhibition of allergic responses in AR patients. Our findings demonstrate a role for pDC in the regulation of allergic reactions in the upper airway and raise the possibility that pDC-targeted therapy might be useful in the management of AR.Q:
